Leading Oncology Drug Developers Join Coiled Therapeutics Medical Advisory Board to Support AO-252 Clinical Development
Coiled Therapeutics (AIM: COIL, OTCQB: COTXF), the clinical-stage oncology company developing precision medicines for hard-to-treat cancers, announces the appointment of Professor Guru Sonpavde, MD and Dr Alexander Spira, MD PhD FACP as founding members of its Medical Advisory Board ("MAB"). Their appointments further the clinical leadership supporting AO-252 as the programme advances through Phase I development following encouraging early clinical signals and follows the recent addition of Professor Ozgur Sahin as Scientific Advisor to the Company.
Professor Guru Sonpavde, MD
Professor Sonpavde is one of the world's foremost clinical investigators in genitourinary oncology and drug development across solid tumours. He currently serves as Director of Genitourinary Oncology and Phase I Clinical Research and holds the Christopher K. Glanz Endowed Chair for Bladder Cancer Research, at the AdventHealth Cancer Institute in Orlando, Florida, where he also holds the rank of Professor of Medicine at the University of Central Florida. He previously served as Bladder Cancer Director at the Dana-Farber Cancer Institute and Associate Professor of Medicine at Harvard Medical School.
Professor Sonpavde is the Principal Investigator or steering committee leader on multiple pivotal Phase III international trials with over 500 peer-reviewed publications. His research encompasses novel therapeutic targets, antibody-drug conjugates, immune checkpoint inhibitors and the translational science underpinning precision oncology. He has co-led landmark trials resulting in approvals including nivolumab plus gemcitabine-cisplatin for advanced urothelial carcinoma (CheckMate 901) and enfortumab vedotin versus chemotherapy (EV-301) and serves on key steering committees evaluating emerging systemic therapies across genitourinary and other solid tumours.
Dr Alexander Spira, MD PhD FACP
Dr Spira is an internationally recognised leader in early-phase oncology drug development and thoracic oncology. He serves as CEO and Director of NEXT Oncology Virginia and as Co-Director of Clinical Research at Virginia Cancer Specialists, where he leads one of the most active phase I programmes in the United States community oncology setting. He also holds an appointment as Assistant Professor of Oncology (Part-Time) at the Johns Hopkins School of Medicine.
Dr Spira has served as Chair of the US Oncology Research Executive Committee, Chair of the ASCO Broadening Clinical Trial Eligibility Committee and as a member of national policy and guideline bodies across ASCO and the Society for Immunotherapy of Cancer. He has contributed as principal investigator or co-investigator to numerous first-in-human and registration-enabling studies spanning targeted therapies, antibody-drug conjugates and immuno-oncology agents across a wide range of solid tumour types, with over 200 peer-reviewed publications. His depth of experience in early-phase trial design makes him a particularly valuable advisor as AO-252 progresses through dose escalation and into targeted expansion cohorts.
As founding MAB members, Professor Sonpavde and Dr Spira will provide strategic clinical guidance on the development of AO-252, including expansion cohort design, indication prioritisation, combination therapy strategies and competitive clinical positioning. Their appointments follow the recent positive clinical update and are structured to accelerate progress from dose escalation into the expansion phase anticipated in H2 2026.

About AO-252
AO-252 is a first-in-class, orally administered, brain-penetrant small molecule inhibitor of Transforming Acidic Coiled-Coil containing protein 3 (TACC3). TACC3 is a validated oncology target that is frequently overexpressed in many aggressive, hard-to-treat solid tumours but is dispensable in normal adult cells, providing a wide therapeutic window.
By selectively disrupting cancer-critical protein-protein interactions at the TACC3 C-terminal domain, AO-252 induces mitotic and replication stress, impairs DNA damage repair, and triggers cancer cell death. Notably, AO-252 has demonstrated the ability to cross the blood-brain barrier, a key differentiator that may enable treatment of both primary brain tumours and brain metastases, areas of significant unmet medical need. Preclinical and early clinical observations support further evaluation of the programme's potential across multiple tumour types.
AO-252 is currently being evaluated in an ongoing Phase I open-label dose-escalation study in the United States. Early clinical observations have demonstrated encouraging anti-tumour activity and a favourable safety profile, with the Company planning to initiate dose expansion cohorts in selected indications, including prostate and ovarian cancer, during 2026.
