Coiled Therapeutics (AIM: COIL, OTCQB: COTFX), the clinical-stage oncology company developing precision medicines for hard-to-treat cancers, is pleased to announce the appointment of Professor Ozgur Sahin, Ph.D. as a Scientific Advisor to the Company. This is the first in a series of planned appointments as Coiled Therapeutics establishes a wider Scientific Advisory Board.

 

Professor Sahin is one of the world's leading authorities on TACC3 biology and a named inventor on the foundational patents underpinning the Company's lead programme, AO-252, a first-in-class precision oncology therapy designed to target a novel cancer vulnerability found in multiple aggressive cancers. He is an experienced cancer biologist with a strong background in cancer biology, functional genomics, bioinformatics and drug discovery. He currently serves as SmartState Endowed Chair and Professor in the Department of Biochemistry & Molecular Biology at the Medical University of South Carolina ("MUSC"), where he also serves as Co-Leader of Cancer Biology and Immunology programme, Program Director for Basic Science Translation and Co-Leader of the Breast Transdisciplinary Cancer Team at the Hollings Cancer Center.

 

Professor Sahin has authored over 80 peer-reviewed publications, received more than 20,000 citations, secured over 20 grants as principal investigator from leading agencies including the US National Institute of Health, and is a co-founder of two oncology drug discovery companies: OncoCube Therapeutics and LoxiGen, Inc. He previously held research positions at the German Cancer Research Center (DKFZ), the University of Texas MD Anderson Cancer Center, Bilkent University and University of South Carolina.

 

In his role as Scientific Advisor, Professor Sahin will work closely with the Coiled Therapeutics leadership team to support AO-252 through a series of important development milestones over the next 12 months, including the transition into targeted dose expansion cohorts, the introduction of the next-generation formulation and the initiation of the combination therapy protocol later this year, where his expertise in immune-modulatory oncology and TACC3 biology will be of particular value.

About AO-252

AO-252 is a first-in-class, orally administered, brain-penetrant small molecule inhibitor of Transforming Acidic Coiled-Coil containing protein 3 (TACC3). TACC3 is a validated oncology target that is frequently overexpressed in many aggressive, hard-to-treat solid tumours but is dispensable in normal adult cells, providing a wide therapeutic window.

 

By selectively disrupting cancer-critical protein-protein interactions at the TACC3 C-terminal domain, AO-252 induces mitotic and replication stress, impairs DNA damage repair, and triggers cancer cell death. Notably, AO-252 has demonstrated the ability to cross the blood-brain barrier, addressing a significant unmet medical need for the treatment of brain metastases.

 

The asset is currently in an ongoing Phase I open-label dose-escalation study and early clinical signals have shown encouraging anti-tumour activity and a benign safety profile, with the Company planning to initiate dose expansion cohorts in lead indications, including prostate and ovarian cancer, during 2026.
