I give below details of changes in interests in the American Depositary Shares
(ADSs) of GlaxoSmithKline plc in respect of the under-mentioned Director:-


Dr T Yamada     Exercise of options on 3 May 2006 over 21,380 ADSs granted on 
                21 November 1996 under the SmithKline Beecham US 1991 Share 
                Option Plan at a price of $28.16 per ADS followed by the sale 
                of the resulting ADSs at a price of $56.77 per ADS.

                The Company was advised of these transactions on 4 May 2006.

This notification relates to a transaction notified in accordance with
Disclosure Rule 3.1.4R(1)(b).
